Upfront Briefing

AstraZeneca put $40M on the table for insitro's AI target hunt in MASH and cardiomyopathy, with $1.46B more riding on programs that don't exist yet. Cheap ticket, long ride. Over at EMA, the CHMP waved through Revolution Medicines' Rasonque for previously treated metastatic pancreatic cancer, so the RAS(ON) story now has two continents.

OrbiMed banked $3B, including its biggest Asia fund yet, so the specialist LPs are still picking up the phone. Paul Stoffels' new Armed Cell picked up his old Galapagos CAR-T portfolio for €8M upfront, City Therapeutics starts trading today after an upsized $200M IPO, and UEG Week is under way in Barcelona with Lilly's Omvoh making the rounds.

  • Healthcare led the S&P sector board as money rotated out of AI and chip names: XLV +1.9%.

  • September PPI (8:30 ET) ran hot at +1.3% m/m and +6.3% y/y vs ~5.5% expected, a day after Wednesday's hot CPI, yet the 10-year fell ~9bp to ~5.10%.

The Big 3

1️⃣ — AstraZeneca, insitro strike up to $1.5B AI drug deal

  • AstraZeneca will pay insitro $40M upfront and up to $1.46B in development and commercial milestones (~$1.5B total) to run insitro's AI target-discovery platform in MASH and cardiomyopathies, with exclusive worldwide rights to advance programs it selects. Target count and royalties were not disclosed.

  • Why it matters: Strip out the headline and AstraZeneca is paying under 3% of the $1.5B for a discovery option; the other $1.46B rides on programs that do not exist yet. That is cheap access to insitro's human-genetics and cell-imaging platform in two areas where AZ needs fresh biology: MASH, and cardiomyopathy after Wainua missed the primary endpoint of the CARDIO-TTRansform ATTR-CM outcomes trial this summer. It extends an AI run at AZ that already includes the Algen immunology pact, the Owkin licence and the Modella AI acquisition. insitro adds a fourth big-pharma partner after Gilead, BMS and Lilly, which validates the platform but says little about what its last disclosed raise, $400M in 2021, is worth today. Nothing here reaches the clinic for years.

2️⃣ — Rasonque wins CHMP backing in pretreated pancreatic cancer

  • EMA's CHMP recommended EU approval of Revolution Medicines' Rasonque (daraxonrasib), an oral RAS(ON) multi-selective inhibitor, as monotherapy for adults with metastatic pancreatic adenocarcinoma after at least one prior systemic therapy. In Phase 3 RASolute 302 (n=500) median OS was 13.2 vs 6.7 months on chemotherapy (HR 0.40) and median PFS 7.2 vs 3.6 months (HR 0.49). The European Commission makes the final call.

  • Why it matters: EU clearance would complete a transatlantic launch that began with FDA approval on 26 August, on a label with no mutation restriction from a trial where 92% of patients carried a RAS G12 mutation. An OS hazard ratio of 0.40 against chemotherapy in second-line PDAC is the bar every next-wave KRAS program now gets measured against, Verastem's VS-7375 included. EMA's own summary cites the more conservative ad hoc OS cut (12.4 vs 6.7 months, HR 0.49), and tolerability gives EU payers leverage: diarrhea 63%, acneiform dermatitis 62%, stomatitis 56%. The bigger value pool is first line, where FDA granted Breakthrough Therapy designation to the chemo combination in September and RASolute 309 with zoldonrasib in G12D began dosing this month.

3️⃣ — OrbiMed closes $3B across Asia and flagship venture funds

  • OrbiMed raised $3B across two vehicles: OrbiMed Asia Partners VI at more than $1B, its largest Asia fund, and OrbiMed Private Investments X at more than $1.9B for U.S. and European life-science venture. Both invest across biopharma, medtech and tech-enabled healthcare services.

  • Why it matters: The Asia number is the signal. Fund VI is roughly a third larger than Fund V's $751M, raised while China-sourced assets take a growing share of big-pharma in-licensing and the COINS Act extension still hangs over those deals. The flagship tells a flatter story: Fund X at $1.9B+ is barely above its $1.86B predecessor, so OrbiMed is scaling Asia faster than its U.S./EU venture book. Add Forbion's €2.3B close on 7 October and TCGX's $600M Asia fund on 8 October and specialist LPs are still writing checks, but into established platforms. For private biotechs that means syndicate leads with dry powder into 2027; for emerging managers it means a harder raise.
