Upfront Briefing
Third time’s the charm: Replimune got Tudriqev past the FDA in advanced melanoma, two rejections and one 10-3 adcomm later, with a $450,000 list price to show for the wait. Latigo picked the same evening to price an upsized $345.6M IPO at the top of its range, which is a fairly loud way of saying the pain window is open. Tarsus, meanwhile, put $450M into Alkeus for a Stargardt drug whose pivotal data lands in 2029 — patience, or a very long-dated call option, depending on your seat. Elsewhere: earnings, guidance raises and a couple of programs quietly taken out back.

- Biotech outran a soft tape — XBI up 1.0%, BBC up 1.7% against a 0.2% S&P decline — as money rotated out of memory and software (Sandisk off 6%, Western Digital 13%, AppLovin nearly 20%) and into defensives, with Moderna’s mFlusiva clearance the prior evening giving the sector a live catalyst.
- The Dow snapped a five-session record run, down 464 points (0.85%), as Brent pushed toward $83 on Iran–Oman talks over reopening the Strait of Hormuz; gold added 1.8% and VIX firmed to 15.27 while the 10-year sat at 4.66%, leaving Friday’s July payrolls as the swing factor after Challenger put July layoffs at a two-year low and claims printed 199,000.
- Market data: U.S. cash close Thursday, 6 August 2026.

FDA approves Replimune's Tudriqev in advanced melanoma
|
- Tudriqev won FDA accelerated approval in advanced melanoma after two earlier rejections and a 10-3 advisory committee vote in favor.
- Why it matters: Accelerated approval rests on a 24.2% ORR and 14.1-month median duration of response among 91 efficacy-evaluable patients — the same single-arm IGNYTE dataset the FDA rejected in July 2025 and again in April 2026. What changed was the 10-3 adcomm, not the data. The $450,000 list price gives Replimune its first product revenue against a going-concern flag, under a year of cash and a proposal to double authorized shares to 300 million; approval also pushes the term-loan maturity from October 2027 to October 2029. IGNYTE-3, roughly 400 patients with OS as primary, reads out in 2027 and can take it all back.

Latigo prices upsized $345.6 million IPO
|
- Latigo priced an upsized $345.6M IPO at $18.00 per share for 19.2M shares to fund its non-opioid pain pipeline.
- Why it matters: Upsizing from 16M shares at $16–18 to 19.2M at $18 is a 27% bump on the $272M target and the third top-of-range biotech pricing this week. The money funds a direct run at Vertex’s Journavx, which cleared in January 2025 and managed roughly $90M in year one — soft enough that Latigo’s pitch is efficacy, not category creation: SPID48 of 62.1 in the high-dose arm of a 343-patient abdominoplasty Phase 2b, published in NEJM. Phase 3 bunionectomy starts this half on the FDA’s preferred pain model, the one Vertex used. Topline 2H27, so nothing resolves before then.

| 3 | Tarsus buys Alkeus for $450M, funds it with a $125M PIPE |
- Tarsus agreed to acquire Alkeus for $450M to secure gildeuretinol, a once-daily oral candidate in Phase 3 for Stargardt disease, alongside an oversubscribed $125M PIPE.
- Why it matters: $270M cash plus $180M in Tarsus stock, with up to $350M more contingent on gildeuretinol reaching market — and the $125M PIPE comes from Bain Capital Life Sciences and TCGX crossing over from Alkeus’s own cap table, which is how you fund a deal without touching the balance sheet. Stargardt has no approved therapy and roughly 36,000 US patients, but Belite Bio’s tinlarebant already won its Phase 3 and is in front of the FDA, so Tarsus is buying second position in a race it doesn’t lead. The asset has also flunked a Phase 3 in geographic atrophy. Pivotal readout is 2029.
