Upfront Briefing

Ultragenyx finally got its Sanfilippo approval — two days early, full approval rather than the accelerated nod it asked for, and a priority review voucher in the bag. Novo wrote a $1.4B biobucks cheque for Orbis’ oral macrocycles without disclosing a single number underneath it, and Roche put $36.5M down on screening 300,000 bispecific combinations.

The financing window is doing its best 2021 impression: two reverse mergers and an upsized IPO in one session, $755M of fresh paper between them. Elsewhere, policy stayed clear as mud, with pharma tariff rules unresolved 12 days before go-live.

  • Biotech led, and it was beta rather than defensives: XBI +2.6%, NBI +2.2% and BBC +2.2% against XLV’s +0.6% and the S&P 500’s +1.1%.
  • The broader bid came from crude and the long end: Brent slipped to about $104 on signs Saudi supply is set to re-flow, and TLT added 1.1% as the 10-year backed off the 2007 high it touched on Wednesday. First session after the Fed’s 25bp hike to 3.75%–4.00%.

The Big 3

  • The FDA granted standard full approval to Fayuvi (rebisufligene etisparvovec-hopf), the AAV9 gene therapy known as UX111, for paediatric MPS IIIA with preserved neurodevelopmental function — two days ahead of the 19 September PDUFA and against a resubmission that had sought accelerated approval. Product ships to qualified treatment centres in 30–60 days.
  • Why it matters: Full approval, not the accelerated nod Ultragenyx asked for, on a resubmission that followed a July 2025 CRL over manufacturing — the Bedford CMC fix cleared inspection in August and this approval is the receipt. The commercial line is thin: 1,500–2,500 US patients and 6,000–8,000 worldwide, so Fayuvi is a validation asset, not a revenue story. The priority review voucher granted alongside it is the nearer-term cash and is separately saleable to a peer. Second gene therapy approval and sixth FDA nod overall for RARE, which answers the platform question that hung over the stock through 2025.

Novo, Orbis strike up to $1.4B oral macrocycle deal
  • Novo Nordisk signed a multi-target discovery and licensing agreement with Denmark’s Orbis Medicines worth up to $1.4B in upfront and milestone payments, plus tiered royalties and an undisclosed strategic equity investment. Neither the targets nor the split between upfront and milestones was disclosed; Orbis’ nGen platform has reached up to 18% oral bioavailability in preclinical work.
  • Why it matters: Read $1.4B as a ceiling, not a cheque. Neither side broke out the upfront, and on a discovery-stage multi-target deal the overwhelming majority of that number sits behind preclinical, clinical and sales gates. What is disclosed matters more: Novo is buying a shot at oral delivery for targets its SNAC peptide technology, acquired with Emisphere in 2020, cannot reach. Novo Holdings seeded Orbis in 2021 and joined the €90M Series A in January 2025, so this is an insider adding, not a contested process; Lilly also sits on the register. Hanmi/Genentech put $190M upfront against $2.31B. This one names no floor at all.

North Immunology takes $180M and a Nasdaq shell at Dupixent
  • ADAR1-incubated North Immunology will go public through an all-stock reverse merger with Aethlon Medical, alongside a $180M oversubscribed private placement — roughly $146M of new money plus about $34M of converting notes. AEMD rose 471%. Lead asset NOR-101, a half-life-extended IL-13 x IL-18 bispecific, enters Phase 1a in Q1 2027; North holders take 95.25% and Aethlon holders 4.75% plus a CVR on the legacy Hemopurifier business. Ticker becomes NRTX.
  • Why it matters: The financing is the story, not the science. This is roughly the twenty-fourth biotech reverse merger of 2026 and it lands the same session Lisata took in Marea with a $225M placement and Electra priced an upsized $350M IPO. Bain Capital Life Sciences, Janus Henderson, Deep Track, Longitude, Soleus, Invus, Farallon, Adage and TCGX all bought paper in a preclinical asset, which is the crossover bid telling you it will fund 2028 data. The comps are Slate/Fulcrum at $245M, Avenzo/Rallybio at $215M and Avere/NextCure at $320M. NOR-101 adds IL-18 blockade to the IL-13 axis Dupixent owns, aimed at the non-type-2 patients biologics miss; no human data until mid-2027.
