  UPFRONT BRIEFING

FDA accepted uniQure's Huntington's gene therapy under accelerated approval with priority review, a 1 May 2027 action date and an adcomm pencilled in. Inovio, meanwhile, finally has an approved product. Voceryq, a DNA medicine for recurrent respiratory papillomatosis, cleared after Friday's close and puts Precigen's Papzimeos on notice.

Lonza is buying five US CDMO sites from Merck KGaA, adding ADC payload-linker and viral-vector capacity, and Karyopharm went into Chapter 11 with Ayrmid lined up as lead bidder. On the calendar: SITC opens tomorrow in Phoenix, AASLD follows Thursday and AHA on Friday.

TAPE ACTION

  • Biotech underperformed: XBI (2.5%), BBC (3.0%) and NBI (1.5%) versus XLV (0.1%). Single names moved both ways: Inovio nearly tripled to $1.89 on Friday's after-hours Voceryq approval while Precigen fell ~26%, and Bright Minds sank on a partial clinical hold disclosed Monday morning.

  • Oil and AI pulled opposite ways: Brent fell 7.6% to $104.77 on signs of a US-Iran diplomatic track, lifting the Dow +0.4%, while an afternoon tech selloff (Marvell (12.7%) and Lumentum (18.6%) on their combination, Oracle (8.8%) on a $10B stock sale) erased an early 1.1% S&P gain. NDX (1.4%), S&P (0.5%), RUT (0.2%); ISM manufacturing 55.5 vs 54.5 expected; 10Y -3.5bp to 4.89% into today's midterms.

THE BIG 3

1️⃣ — uniQure's Huntington's gene therapy gets a 1 May PDUFA and an adcomm

  • FDA accepted the BLA for ifezuntirgene inilparvovec (AMT-130) under accelerated approval with priority review, PDUFA 1 May 2027; the acceptance letter says FDA tentatively plans an advisory committee, date not set. The filing rests on 36-month Phase 1/2 data (51 patients dosed) versus a propensity-matched Enroll-HD external control; confirmatory trial ~200 patients, 2:1 vs standard of care, Total Functional Capacity at 36 months, US screening by year end.

  • Why it matters: An external-control filing gets argued in public. The BLA leans on 36-month Phase 1/2 data against a propensity-matched Enroll-HD comparator rather than a randomized arm, which is the evidence an advisory panel tends to pull apart: how patients were matched, who dropped out, and whether slowing on a composite scale is a functional benefit. Priority review on top of Breakthrough, RMAT and Fast Track tags says FDA wants to move; the planned adcomm says it wants cover. The agreed ~200-patient randomized confirmatory trial, with a 36-month TFC endpoint, matters as much as the date, because accelerated approvals now come with withdrawal risk. A clean panel would give other CNS gene therapies built on natural-history controls a template; a split vote reprices QURE into May.

2️⃣ — Inovio's Voceryq wins accelerated approval in RRP, Precigen takes the hit

  • FDA granted accelerated approval to Voceryq, a DNA immunotherapy given with the CELLECTRA 5PSP device, to reduce surgical papilloma excisions in adults with recurrent respiratory papillomatosis (approved Fri 30 Oct, after the close). In RRP-001, 32 adults enrolled and 26 were in the efficacy population (3+ surgeries in the prior 12 months, median 4.5); complete response, meaning no excision through 12 months, was 7/26 (27%, 95% CI 12-48%). Product ships in ~60 days, full launch Q1 2027; confirmatory protocol due by year end.

  • Why it matters: A small label with a large read-through. Voceryq is Inovio's first approved product after years of DNA-platform setbacks, and the stock nearly tripled to $1.89 on Monday. The loser is Precigen: Papzimeos has had the RRP market to itself since its August 2025 approval and booked $74.7M of net sales in 1H26, most of the company's revenue, and PGEN fell ~26%. On paper the data gap is wide, a 27% complete response from 26 patients in a single-arm study. The commercial fight is about convenience, payer appetite for a second option and Inovio's ability to fund a launch from a standing start in Q1 2027. The confirmatory protocol, due by year end, is the regulatory overhang.

3️⃣ — Lonza buys five US CDMO sites from Merck KGaA

  • Lonza signed a definitive agreement to buy five US CDMO sites from Merck KGaA, Darmstadt, at an enterprise value in the low- to mid-triple-digit millions of Swiss francs, in cash from balance sheet and existing facilities; close expected mid-2027. The sites are St. Louis (bioconjugates), Madison and Verona, WI (high-potency payload-linkers), Carlsbad, CA (viral vectors, aseptic fill-finish) and Indianapolis (LNPs, liposomes, PEGylated proteins), with ~1,100 staff; Lonza's US sites go from four to nine, and ~CHF250M of 2028 sales is expected.

  • Why it matters: This is a supply bet on antibody-drug conjugates. St. Louis and the two Wisconsin sites cover conjugation and high-potency payload-linkers, the capacity-constrained steps in ADC manufacturing. Carlsbad and Indianapolis add viral-vector and LNP work, a softer demand pocket after the gene-therapy pullback, which is why Lonza guides to a sub-group core EBITDA margin on ~CHF250M of 2028 sales and only reaches group margin by the mid-2030s. A price in the low-to-mid hundreds of millions of francs is modest for five running US sites and 1,100 staff. Lonza kept its 2026 outlook, buyback and BBB+ rating, so this is a bolt-on, not a stretch. Merck KGaA, for its part, exits US contract manufacturing.
