Upfront Briefing
Vaxcyte posted the kind of Phase 3 print that rewrites a cap table, then did the grown-up thing and launched a $1B raise before the confetti settled. Shionogi paid $2B upfront for IntraBio and a two-label orphan drug that sold under $70M last year, a reminder that commercial rare disease still clears at a premium. Genentech handed Alector $100M upfront for a preclinical Parkinson's enzyme, which reads as much like a runway extension as a neuro bet. On the calendar: Cell & Gene Meeting on the Mesa wraps Wednesday in Phoenix, and CPhI Europe opens in Milan today.

- Biotech outran the broad market: XBI and NBI each rose 1.1% against 0.7% for XLV, with BBC up 0.1%. The group's biggest single-name catalyst landed pre-open, with Vaxcyte's OPUS-1 topline out ahead of its 8:00 a.m. ET call and PCVX trading up as much as ~50%; its $1B financing launched at 4:28 p.m. ET, after the bell.
- Tech led: the Nasdaq 100 rose 0.9% to a record and the S&P 500 0.7%, even as the 10-year yield climbed about 4bp to ~5.32%, a 52-week high, after September ISM services printed 55.9 vs 55.7 expected with prices paid at 74. Minutes from the Fed's September meeting land Wednesday.
- Market data: U.S. cash close Monday, 5 October 2026.

Vaxcyte's VAX-31 holds its own against Prevnar 20 and Capvaxive in pivotal OPUS-1
|
- OPUS-1 (n=4,047) met its primary immunogenicity endpoints. In adults 50+, VAX-31 was non-inferior on all 20 serotypes shared with Pfizer's PCV20 and on 17 of 19 shared with Merck's PCV21 at the standard bound (serotypes 3 and 12F cleared the historical 0.5 bar), and superior on four. OPUS-2 and OPUS-3 read out in 1H27; the BLA is planned for 1H28.
- Why it matters: OPUS-1 is the readout the adult pneumococcal market was waiting for: a 31-valent shot that concedes almost nothing to Prevnar 20 or Capvaxive on shared serotypes and adds coverage neither offers. The bear case is serotypes 3 and 12F missing the strict bound against PCV21, but both cleared the historical 0.5 bar, and Mizuho called the package near best case. Prevnar is a $6B-plus franchise and Capvaxive is Merck's wedge into it, so the debate moves from approvability to share. The catch is timing: OPUS-2 and OPUS-3 read out in 1H27 and the BLA is not planned until 1H28, which is why management launched a $1B stock-and-convert raise into the spike. Launch capex arrives well before revenue.

Shionogi to acquire IntraBio for $2B
|
- Shionogi will pay $2B upfront for privately held IntraBio and its oral drug Aqneursa (levacetylleucine), approved for Niemann-Pick disease type C in September 2024 and for ataxia-telangiectasia in late September. Closing is expected in November–December.
- Why it matters: Shionogi is paying roughly 29x trailing sales: IntraBio booked a little under $70M in net sales last year, per Shionogi's deal materials. The premium is for the A-T label, where Aqneursa is the first approved therapy, plus a Phase 3 in CACNA1A disorders; in Niemann-Pick type C it shares the market with Zevra's Miplyffa. This is Shionogi's third acquisition in about a year, after Japan Tobacco's pharma unit for $1B and Tanabe's Radicava portfolio for $2.5B, and the pattern is consistent: buy marketed or near-marketed specialty revenue rather than discovery. A private, commercial orphan company clearing $2B upfront is a live comp for late-stage rare-disease boards weighing an IPO against a sale.

Alector lands $100M upfront from Genentech for preclinical Parkinson's enzyme
|
- Genentech takes exclusive global rights to AL050, a GCase enzyme replacement therapy carried across the blood-brain barrier on Alector's ABC platform, for GBA1-linked Parkinson's: $100M upfront, up to $1.17B in milestones and tiered royalties. Alector keeps the platform; its shares rose more than 20%.
- Why it matters: $100M upfront for an asset still in IND-enabling work is a full price, and it extends Alector's cash runway into 2029 after GSK and AbbVie both exited earlier partnerships. Roche is buying the carrier as much as the enzyme: it already runs its own Brainshuttle platform behind trontinemab in Alzheimer's, so licensing a second blood-brain barrier technology buys delivery options in neurology. The biology risk is real. GBA1 variants are the most common genetic risk factor for Parkinson's, but no GCase-directed therapy has yet shown clinical benefit, and AL050 has no human data. ALEC now trades on the ABC platform and three wholly owned programs, AL137, AL164 and AL062, with the Genentech milestones as upside on top.
