Upfront Briefing
BioNTech's off-the-shelf mRNA cancer vaccine BNT113 whiffed on progression-free survival in its Phase 3 head-and-neck test, leaving overall survival to carry the whole trial. That makes three mRNA-oncology stumbles for BioNTech this year, and Moderna and Merck's intismeran just inherited a little more of the category's burden of proof. Dealmakers kept swiping anyway. Roche went fat-hunting in China for the seventh time this year, licensing Rona's ALK7 RNAi programme for up to $100M upfront and near-term (~$1.16B all-in), while Pfizer inked a $500M+ Sec61 pact with Paris-based Enodia a week after Lilly doubled down on Gate. World ADC wraps in San Diego tomorrow.

- Biotech led healthcare lower, with XBI (3.0%), NBI (2.4%) and BBC (5.6%) all trailing XLV (1.4%) and no single sector catalyst. Among pre-open single-name losers, J&J fell about 5% intraday on a Q3 MedTech miss and $5.5B talc charge, BioNTech slid on its BNT113 Phase 3 PFS miss and Syndax dropped about 17% after ending Niktimvo in IPF.
- The S&P 500 finished flat and the Nasdaq 100 fell 0.3%: memory names (Micron, Seagate, Western Digital) sold off on reports China's CXMT is moving to next-generation DRAM, and bank earnings split (JPMorgan miss, Goldman beat). Energy and utilities led as Brent slid about 2% toward $104.5 on eased Russian diesel curbs, and Treasury yields drifted lower into Wednesday's 8:30 ET September CPI print.

BioNTech's BNT113 misses PFS in Phase 3 head-and-neck test
|
- BNT113, an off-the-shelf mRNA immunotherapy encoding HPV16 E6/E7 antigens, failed to significantly improve progression-free survival when added to Keytruda in the Phase 3 portion of AHEAD-MERIT (n=358), in first-line metastatic HPV16-positive head and neck squamous cell carcinoma. No hazard ratio was disclosed. The IDMC recommended the trial continue to its overall survival readout, the other dual primary endpoint. BNTX opened about 5% lower.
- Why it matters: PFS and OS were dual primaries, so AHEAD-MERIT can still win on survival, but BMO now holds that outcome with "significantly" less confidence. The bull case is Keytruda's own Phase 3 head-and-neck precedent, which missed PFS and later won on OS; the base rate for that conversion is poor. This is BioNTech's third mRNA-oncology setback of 2026, after autogene cevumeran was dropped in bladder cancer and halted for futility in colorectal cancer, leaving both its fixed-antigen and individualized approaches unproven in randomized settings. Moderna, whose intismeran data with Merck lifted the group in August, slipped about 3% premarket. With no HR and no OS timing, BNT113 is now a lottery ticket rather than a value driver.

Roche licenses Rona's ALK7 RNAi obesity drug for up to $1.16B
|
- Roche took an exclusive global licence to China-based Rona Therapeutics' ALK7-targeting RNAi obesity candidate, built on Rona's extrahepatic delivery platform, for up to $100M in upfront and near-term payments, up to $1.06B in milestones (~$1.16B total) and tiered royalties. Asset name and stage were not disclosed. Endpoints counts it as Roche's seventh China-linked deal of 2026.
- Why it matters: ALK7 is the fat-first, muscle-sparing bet: the receptor sits mainly on adipocytes, carriers of variants have lower waist-to-hip ratios, and the pitch is visceral-fat loss stacked on an incretin. Roche is arriving late. Arrowhead and Alnylam are already in Phase 1, Arrowhead showed a 14.1% placebo-adjusted visceral-fat cut eight weeks after one dose, GSK paid SiranBio $55M for a Phase 1 asset in May, and Novo says it has several ALK7 programs. With petrelintide's glycemic data underwhelming and a muscle-sparing antibody returned after interim futility, Roche needs shots beyond Carmot's incretins. The $100M is modest for an undisclosed-stage asset, and China origin carries the usual COINS Act overhang.

| 3 | Pfizer joins the Sec61 race with $500M+ Enodia pact |
- Pfizer signed a multi-target discovery collaboration with Paris-based Enodia Therapeutics on small-molecule Sec61 inhibitors, which stop disease-driving proteins at the point of synthesis and route them to degradation. The upfront is undisclosed; Fierce and BioSpace put potential value above $500M plus tiered royalties. Enodia discovers and optimizes candidates; Pfizer takes clinical, manufacturing, regulatory and commercial work.
- Why it matters: Sec61 is the channel that threads new secretory and membrane proteins into the ER, so blocking it knocks down cell-surface and secreted targets without needing the ligandable pocket a PROTAC or molecular glue requires. Pfizer is not a stranger here: Pfizer Ventures co-led Enodia's €20.7M seed in January. The timing tracks Lilly's second Gate Bioscience Sec61 pact on 5 October (>$870M in biobucks). The open question is selectivity, since Sec61 is housekeeping machinery; the main human datapoint is Kezar's Phase 1 showing tolerable Sec61 inhibition in solid tumors, and Enodia bought preclinical Sec61 assets from Kezar in March for $1M upfront. With discovery-stage targets and no upfront disclosed, the headline is mostly back-ended milestones.
